Right target, but missing the bulls-eye for Alzheimer's

Jan. 23, 2013 ? Alzheimer's disease is the most common cause of late-life dementia. The disorder is thought to be caused by a protein known as the amyloid-beta protein, or Abeta, which clumps together in the brain, forming plaques that are thought to destroy neurons. This destruction starts early, too, and can presage clinical signs of the disease by up to 20 years.

For decades now, researchers have been trying, with limited success, to develop drugs that prevent this clumping. Such drugs require a "target" -- a structure they can bind to, thereby preventing the toxic actions of Abeta.

Now, a new study out of UCLA suggests that while researchers may have the right target in Abeta, they may be missing the bull's-eye. Reporting in the Jan. 23 issue of the Journal of Molecular Biology, UCLA neurology professor David Teplow and colleagues focused on a particular segment of a toxic form of Abeta and discovered a unique hairpin-like structure that facilitates clumping.

"Every 68 seconds, someone in this country is diagnosed with Alzheimer's," said Teplow, the study's senior author and principal investigator of the NIH-sponsored Alzheimer's Disease Research Center at UCLA. "Alzheimer's disease is the only one of the top 10 causes of death in America that cannot be prevented, cured or even slowed down once it begins. Most of the drugs that have been developed have either failed or only provide modest improvement of the symptoms. So finding a better pathway for these potential therapeutics is critical."

The Abeta protein is composed of a sequence of amino acids, much like "a pearl necklace composed of 20 different combinations of different colors of pearl," Teplow said. One form of Abeta, Abeta40, has 40 amino acids, while a second form, Abeta42, has two extra amino acids at one end. Abeta42 has long been thought to be the toxic form of Abeta, but until now, no one has understood how the simple addition of two amino acids made it so much more toxic then Abeta40.

In his lab, Teplow and his colleagues used computer simulations in which they looked at the structure of the Abeta proteins in a virtual world. The researchers first created a virtual Abeta peptide that only contained the last 12 amino acids of the entire 42-amino-acid-long Abeta42 protein. Then, said Teplow, "we just let the molecule move around in a virtual world, letting the laws of physics determine how each atom of the peptide was attracted to or repulsed by other atoms."

By taking thousands of snapshots of the various molecular structures the peptides created, the researchers determined which structures formed more frequently than others. From those, they then physically created mutant Abeta peptides using chemical synthesis.

"We studied these mutant peptides and found that the structure that made Abeta42 Abeta42 was a hairpin-like turn at the very end of the peptide of the whole Abeta protein," Teplow said.

The hairpin turn structure was not previously known in the detail revealed by the researchers, "so we feel our experiments were novel," he said. "Our lab is the first to show that it is this specific turn that accounts for the special ability of Abeta42 to aggregate into clumps that we think kills neurons. Abeta40, the Abeta protein with two less amino acids at the end of the protein, did not do the same thing."

Hopefully, the work of the Teplow laboratory presents what may the most relevant target yet for the development of drugs to fight Alzheimer's disease, the researchers said.

Other authors on the study included Robin Roychaudhuri, Mingfeng Yang, Atul Deshpande, Gregory M. Cole and Sally Frautschy, all of UCLA, and Aleksey Lomakin and George B. Benedek of the Massachusetts Institute of Technology.

Funding for the study was provided by grants from the State of California Alzheimer's Disease Research Fund, a UCLA Faculty Research Grant, the National Institutes of Health (AG027818, NS038328) and the James Easton Consortium for Alzheimer's Drug Discovery and Biomarkers.

Computer program finds medieval memes

A new computer program automatically determines the dates of documents from the Middle Ages. It works by looking for words and phrases that were fashionable at the time ? the Ermahgerds of the era ? to determine that a given document is so 1240s, for example.?

The program analyzes property deeds, called charters, written in England between the late 1000s and the mid-1400s. The charters provide a major resource for historians piecing together English history of the time period, but people didn't generally put dates on these documents until after the 1300s.?

The program helps date otherwise mysterious charters, said Gelila Tilahun, a statistician at the University of Toronto who built the program while earning her doctoral degree. Plus, it IDs the memes of the times.

Tilahun hadn't even heard of the charters, which are written in Latin, before she started the project. She still isn't exactly an expert, she says. "I don't know any Latin or, you know, very little medieval history, actually," she told TechNewsDaily. But her program doesn't require her to read Latin or know history. Instead, Tilahun, her doctoral advisor and a University of Toronto historian gave the program a "training set" consisting of the digital files of 326 already-dated charters. (Historians have given many originally undated charters dates by looking for distinctive handwriting or mentions of names or current events.) The program scanned through the training charters, automatically identifying phrases that appeared frequently during certain time periods.?

It then used the patterns it learned to guess years for digital files of other charters the researchers supplied. To test the program, Tilahun and her colleagues gave it a set of charters with known dates, finding that the program's guesses matched.

As a side benefit, the program lists the fleetingly popular phrases it finds. So what were some of the hot sayings of the Middle Ages? "Amicorum meorum vivorum et mortuorum" was common between 1150 and 1240. It's Latin for "of my friends, living and dead." Meanwhile, "Francis et Anglicis," a term of address meaning "to French and English," was used until 1204, when the British lost Normandy to the French.?

The program won't replace historians, though, Tilahun said. It may miss some clues that historians find obvious. For example, if a charter has just one out-of-place word ? "an extreme example would be 'iPad,'" Tilahun said ? the program would ignore the incongruity, but a historian would catch it. Of course, the appearance of iPads in supposedly medieval charters would be easy for anybody to catch, but other words, names or similar clues would require expertise to find.

Tilahun and her colleagues are now trying to train their program to identify the regions charters come from, based on differences between, say, the preferred phrasings of Londoners and Devonshire-dwellers. The researchers are also interested in identifying forgeries by spotting phrases that wouldn't have occurred in a document's supposed year of origin.?

Tilahun has some ideas that are farther afield than these old documents. She's looking to apply a statistical program for identifying bits of DNA that are responsible for turning certain genes on or off. It'll work much like her charters program, she said. "Essentially, it's going from text analysis to analyzing the text of, or the grammar of, our genes," she said.?

Tilahun and her colleagues published their work in the December 2012 issue of the Journal of Applied Statistics. Their paper also appears in arXiv, a free repository of math and physics papers, and was highlighted in MIT Technology Review's arXiv blog.

HSBC to pay $249M to end foreclosure reviews

HSBC Holdings Plc agreed to pay $249 million to end a case-by-case review of past home foreclosures in the United States, bringing the total payout by banks to resolve related issues to $9.3 billion.

London-based HSBC agreed to pay $96 million to eligible borrowers who lost their homes to foreclosure in 2009 and 2010, and provide $153 million in other assistance, including loan modifications and forgiveness.

HSBC said in a statement it was pleased to have reached the agreement and expects to record a pre-tax charge of $96 million in the fourth quarter of 2012 for the cash portion of the settlement. The bank said it expected to cover the loan assistance through existing reserves.

The settlement, with the Office of the Comptroller of the Currency and the Federal Reserve Board, is the 13th the agencies have reached this month.

They stem from reviews of individual loan files the regulators ordered in 2011 and 2012, after widespread mistakes were discovered in the way mortgage servicers had processed home seizures.

The reviews, initially expected to determine which borrowers were harmed and to compensate them based on their individual experiences, proved slow and expensive.

Ten banks, including Bank of America, Wells Fargo, Citigroup, and JPMorgan Chase, agreed to pay a total of $8.5 billion - some in cash, and the rest in loan assistance - to end the reviews last week.

On Wednesday, Goldman Sachs and Morgan Stanley agreed to a similar $557 million deal.

Around 112,000 borrowers whose homes were in foreclosure with HSBC Bank and other HSBC subsidiaries will receive some cash, regulators said.

Regulators said last week the payouts will be based on whether a borrower falls into one of 11 categories. The categories include whether the person was eligible for protections under the Servicemembers Civil Relief Act, whether the borrower was not in default, or whether he or she was denied a loan modification.

The Fed and OCC are expected to reach similar agreements with other servicers that had been asked to conduct the reviews, including Ally Financial Inc, EverBank Financial Corp and OneWest Bank FSB.

Quinton "Rampage" Jackson will fight Glover Teixiera at UFC on Fox 6. If Jackson stays true to his word, it will be his last fight in the UFC. He has complained about being a part of the UFC time and time again. He reiterated on a pre-fight press call that he is done, done, done with the UFC. This is the last fight of his contract, and he has no plans to renegotiate with them.

"I think the UFC don't know how to treat the athletes, in my opinion," he said. "I feel like the fighters do a lot for the sport and I feel like we're not taken care of well enough. I feel like they're getting rich off all of us."

His cries are starting to sound like the lyrics to a Taylor Swift song. Jackson and the UFC are never ever ever getting back together. But if he doesn't fight for the UFC, his options for high-level MMA are limited. He said he could try other combat sports like kickboxing or boxing. Bellator could be an option, but one of Jackson's beefs with the UFC has been their matchmaking. The tournament set-up could be tough for him to deal with.

Either way, enjoy Jackson in the UFC while you can, because it's over for real this time.

Drugs for diabetes? Scientists test the power of plants

Jan. 16, 2013 ? New drugs to treat diabetes are being developed by scientists at the University of Greenwich.

A group of researchers from the university's School of Science, led by Dr Solomon Habtemariam, believe they have identified potential sources of medicines derived from plants which may have fewer adverse side-effects for diabetes sufferers.

The scientists are investigating the properties of two plants found in south-east Asia which they think could have properties that are not only anti-diabetic, but also lipid- or fat-lowering, and so can help tackle obesity.

Dr Habtemariam, a leading expert on drug discovery researches from natural sources, says the work could prove a crucial breakthrough in the treatment of diabetes, which he describes a "growing global epidemic."

"Diabetes is a huge burden to society in general. The search for treatments is making the NHS bankrupt, and this problem is likely to get worse in the next decade. There is no known drug of cure and so, all in all, it's a huge incentive for us to carry out research in this field," he says.

The disease, a result of chronically high levels of glucose in the blood, affects more than 300 million people in the world. It is split into two main classes: type I and type 2. The former normally affects children, while type 2, the most common type, is often diagnosed later in life and in some cases can be managed by diet, exercise and weight loss.

The researchers at Greenwich aim to isolate and identify certain extracts from the plants Cassia auriculata and Cassia alata, which could have 'active ingredients' for treating diabetes. They discovered that one of the compounds isolated from the plant, kaempferol 3-O-rutinoside, has proved to be more than eight times more potent than the standard anti-diabetic drug, acarbose.

The team also found the plants have anti-oxidant properties, which is beneficial when treating diabetes.

"Our other most interesting finding is that many of the active ingredients from the Cassia auriculata plant work through a process called 'synergism' -- in other words, they work together to produce an effect greater than the sum of their individual effects," Dr Habtemariam says. "Overall, this suggests that the crude plant extract has lots of potential to be used clinically for treating diabetes and associated diseases."

The research is ongoing and requires further study and validation, but Dr Habtemariam says the university's School of Science is an ideal place to be conducting his work. "We have both the facilities and the expertise to carry out this research: to isolate chemicals of biological interest, and then to identify what they are. We are only at the drug discovery stage but moving to the clinical trial stage is a very definite goal." Cassia auriculata and Cassia alata grow in a tropical climate. They are popular both as ornamental plants and for their medicinal uses.

Last year Dr Habtemariam led an international research project which revealed the potential of tansy, a flowering plant found in Europe and Asia, as a treatment for the sexually transmitted disease herpes.

Apple's retail executives continue to abandon ship at an alarming rate. Late Tuesday, Apple confirmed that retail VP Jerry McDougal has announced his resignation from Apple.

The departure is, by all means, voluntary and amicable. McDougal says he simply wants to spend more time with his family. Unfortunately for Apple, McDougal had been on a short list of executives to replace John Browett, who exited the position of Apple's retail chief last October.

Apple spokesman Steve Dowling tells All Things D that Apple's VP of Finance Jim Bean will replace McDougal for the time being.

?Retail has an incredibly strong network of leaders at the store and regional level, and they will continue the excellent work they?ve done over the past decade to revolutionize retailing with unique, innovative services and a focus on the customer that is second to none," Dowling says. "Jim Bean is moving to Retail to help support our store teams. Jim has been at Apple for 15 years and is a great leader who understands our culture and focus on customer service.?

Initial reaction to the Bean promotion has been positive.

John Paczkowski of All Things D says "Bean has exactly the sort of operational acumen that would lend itself to the position McDougal?s departure leaves open."

Bibliotech: Texas to unveil nation's first book-less library

A book-less library.

It sounds like an oxymoron, but come the fall of 2013, San Antonio's Bexar County is going to be home to the BiblioTech, the country's first book-less public library. Of course, there will be books -- just e-books, not physical books.

The 4,989 square-foot space will look like a modern library, Bexar County Judge Nelson Wolff, who was inspired to pursue the project after reading Walter Issacson's Steve Jobs biography, told ABC News. (A glance at the photo shows that its inspired by Apple in more ways than one.) Instead of aisles and aisles of books there will be aisles and aisles of computers and gadgets. At the start, it will have 100 e-readers available for circulation and to take out, and then 50 e-readers for children, 50 computer stations, 25 laptops and 25 tablets on site.

"We all know the world is changing. I am an avid book reader. I read hardcover books, I have a collection of 1,000 first editions. Books are important to me," Wolff told ABC News. "But the world is changing and this is the best, most effective way to bring services to our community."

Library goers will be able to take out books on any of the devices in the library, take out one of the 50 e-readers for a period of time or bring their own e-readers to the library and load books onto their own devices. The library will also be partnering with e-book providers or distributors to provide access to over 10,000 titles. The hope is to add to that collection annually. The county is still figuring out who will provide the equipment and has requests for proposals out for the e-readers and other equipment.

Currently Amazon and Barnes & Noble offer services on their respective e-readers which work at libraries. You can take out books on the e-reader devices and then check them back in to the library remotely.

"You will be able to check out a book, read it on-site. It will be a learning environment -- you'll be able to learn about technology itself as well as access a tremendous amount of information," Wolff said. There will also be a children's area with interactive tables and interactive walls. Wolff also said the library is exploring adding other media to the library, like movies and music.

"That could be a real possibility. This is a new venture -- we are starting with the basics -- but we will have the opportunity to add on to that," he said in a phone interview.

But it won't be a completely paper-free library. "The only thing I believe we will charge for is if you want to print out something," Wolff said. "We will charge for the copies you would want to print out."

NBA blogger sees 'clear path' for Kings to Seattle, but nothing's confirmed

There was plenty of basketball blog chatter on Monday about deals to sell the Sacramento Kings, but no confirmation.

Yahoo Sports' Adrian Wojnarowski reported that the NBA discussed deal points on the proposed sale of majority ownership of the Kings to a Seattle-based group during a conference call.

Meanwhile, NBA.com reporter David Aldridge quoted an unnamed industry source saying of the Kings, ?They?ve got a clear path? to Seattle.

?There is never any joy in writing about a team that's about to break a city's heart by leaving,? Aldridge wrote Monday. ?But the truth is the truth, and unless there's some kind of amazing change of heart or desire, the Kings are leaving Sacramento for Seattle. It's just a matter of when the announcement comes.?

But the announcement has yet to come.

These rumors have kept basketball fans on the edges of their seats for almost a week, but no one has found a source close to the proposed dealmakers willing to speak on record.

A spokesman for San Francisco hedge fund manager Chris Hansen ? who wants to build an NBA arena in Seattle and is said to be part of the deal with the Kings' majority owners, the Maloof family ? declined to comment on the situation.

Aaron Pickus, spokesman for Seattle Mayor Mike McGinn, said City Hall assumes that Hansen is working to acquire a team, "but that is not something the city is directly involved in."

?We?re watching the rumors and stories fly around just like everyone else,? Pickus said

The deal reported by Wojnarowski would have the Kings sold to Hansen and Microsoft CEO Steve Ballmer. The group would have until March to file for relocation to bring the team north for the 2013-14 season.

Mario Chalmers' teammates were yelling at him again, as were a few coaches. Dwyane Wade threw a towel his way, and LeBron James even placed the Miami Heat point guard in a headlock.

This was all good.

Chalmers put on a shooting display to remember in Sacramento on Saturday night, making 10 3-pointers ? the most in the NBA this season and good enough to tie a 20-year-old Heat franchise record, part of his career-best 34-point effort as Miami topped the Kings 128-99, the biggest point total put up so far this season by the reigning NBA champions.

"Rio had it goin', man," James said. "And we kept finding him. It was awesome."

Chalmers' final numbers: 12-for-16 from the floor, 10-for-13 from beyond the arc, and one of those misses was a 45-footer that bounced off the rim just before the halftime buzzer. He's long been the player that teammates love to chastise on the floor when things aren't going right for the Heat, though moments like that have become increasingly rare this season.

And on Saturday, no one could have thought of any complaints, anyway.

"I got started early," said Chalmers, who had made 10 of his last 33 tries from 3-point range before his barrage against the Kings. "I started off with two easy layups, and then my teammates did a good job of finding me open behind the 3."

Sure enough, Chalmers' first two baskets Saturday were at the rim, and then everything else was from long range.

For the Heat, it was a bounce-back game after two straight losses.

For Chalmers, it was a bounce-back effort after a chance to be a hero in Portland one game earlier went awry.

The Heat trailed by two with 10.4 seconds left Thursday night against the Trail Blazers, and on a final possession where Wade, James and Chris Bosh all touched the ball, the last shot went to Chalmers ? wide open from the right wing. His 3-pointer looked so good that Wade prematurely started to extend his arms in celebration, but the shot hit the back of the rim, then the front, then bounced away as time expired.

Chalmers walked off the floor, his head down.

One game later, he was back, and in a big way.

"I just wanted to get back into a good rhythm," said Chalmers, who is still perhaps best identified with hitting the overtime-forcing 3-pointer in the NCAA title game where his Kansas team topped Derrick Rose and Memphis for the national championship in 2008. "I feel like I've been struggling all season. Especially missing that last shot in Portland, I wanted to get my mind on things, come out here and redeem myself a little bit."

Miami plays at Utah on Monday night.

Chalmers became the fourth Heat player to score at least 34 points in a game this season ? no other team in the league has more than three who can say that, and eight teams haven't had a single player reach 34 entering Sunday's play. And according to STATS LLC, Chalmers became just the third player since 1990 to make 10 3-pointers on 13 attempts or less in a game.

Ty Lawson was 10-for-11 for Denver against Minnesota on April 9, 2011, and George McCloud went 10-for-12 from long range for Dallas against Phoenix on Dec. 16, 1995.

The only other Heat player with 10 3-pointers in a game was Brian Shaw in 1993.

"This is a classic one for Mario," Wade said. "I liked it because he just had an opportunity in the last game to hit a game-winner and he missed it. I liked that he came back aggressive. Credit to him for tying a Heat record, hitting 10 3's, I mean, that's just a phenomenal feat. We all know that Mario, no matter what goes on, he's a big-game type player."

Heat coach Erik Spoelstra said there was some thought of giving Chalmers a chance to break the team record and try for an 11th 3-pointer. But his 10th came with 3:12 remaining in what was then a 33-point game, and Chalmers was subbed out for the remainder of play 20 seconds later.

With that, the celebrating on the bench began.

"Everything came within the flow," Spoelstra said. "Obviously, the last one, he was going for it, but everybody was rooting him on. It came from a lot of good ball movement, a lot of great spacing, guys were very unselfish and there were a lot of wide-open shots because of that. And it was great to see him knock those down."

Kia's rising star Schreyer to take charge of Hyundai group car design

SEOUL (Reuters) - German designer Peter Schreyer, known as the creative brain behind the Audi TT, will oversee both Kia and Hyundai's designs, the Hyundai automotive group said on Sunday, as the carmaker seeks to outdo German rivals Volkswagen and BMW.

Schreyer's star is rising fast at the South Korean company, where only three weeks ago he was named a president at Kia, the first foreigner to hold the post. He was promoted from chief design officer and executive vice president at Kia.

He will assume the newly created role to oversee the design of both Hyundai and Kia, which together make up the world's fifth biggest automaker. The design operations of the two carmakers had been run by separate design chiefs.

Kia, acquired by Hyundai Motor in 1998 during the Asian financial crisis, has been expanding sales at a faster pace than Hyundai Motor in markets such as the United States and Europe.

Schreyer's new appointments coincide with Hyundai Motor's efforts to move up market and boost margins.

A Hyundai spokesman on Sunday cited the group's need "to more clearly set up each automaker's own, unique brand color".

"President Peter Schreyer, with abundant design experience at world-renowned automakers and deep comprehension of Hyundai-Kia's corporate culture, will play a great role in consolidating the foundation for Hyundai Motor and Kia Motors to take a leap towards a top-class automaker," the spokesman, who was not named, said in a statement.

The promotion is a clear sign of the automotive group's ambition to outdo German rivals Volkswagen AG and BMW in design - a key part of a strategy that has helped the South Korean duo ramp up sales and gain market share even during the global economic downturn in 2009.

Schreyer joined Kia from Volkswagen in 2006.

Born in Germany in 1953, the former Audi designer was reportedly brought in by then Kia president Chung Eui-sun - the only son and heir apparent of the 74-year-old Hyundai Motor Chairman Chung Mong-koo.

Before joining Kia, Schreyer thought Kia's designs were "neutral." He went on to create a "tiger-nose" grille for Kia, and in one stroke, gave a distinct face to Kia models ranging from the small Picanto to the Forte and Optima sedans.

Industrial data showed on Sunday that last year's combined sales by Hyundai and Kia in Germany stood at nearly 155,700 units which made the duo the top exporter of automobiles in the biggest European market.

The Hyundai-Kia team sold a total of 7.12 million vehicles worldwide in 2012, up 8 percent from the previous year and more than their original goal of 7 million. It has said the duo's global sales target for 2013 is a combined 7.41 million.

Manchester scientists have developed a tiny molecular machine that mirrors the function of the ribosome, which builds the proteins in our body's cells.

Just a few millionths of a millimetre in size, the minute machine resembles a ring threaded on a rod.

As this ring moves along the rod, it picks chemical units and assembles them into chains, just as ribosomes join up the building blocks of proteins.

The ultimate goal is to synthesize new drug molecules or new types of plastic.

"Just as robots are used to assemble cars in factories in the big world, one day we hope we will be able to use artificial machines like these in molecular factories to construct new things with great efficiency," said Prof David Leigh FRS from the school of chemistry at Manchester University, UK.

"Ribosomes make proteins, which are just one type of polymer used by nature. In fact, all of biology is based on just four sorts of so-called information polymers - proteins, DNA, RNA and also carbohydrates.

"But with our artificial machines, we're not limited by the same building blocks of nature. So, we should be able to make new materials with other types of building blocks - new types of plastics, new types of catalysts, pharmaceuticals and so on," he told the BBC World Service Science In Action programme.

A scholarly paper in this week's Science Magazine carries details of the research.

The ribosome is one of nature's true marvels. This giant molecule concerns itself with the business of translating and acting on our genetic code.

That code, held in the cell's DNA, is delivered to the ribosome by the "messenger" molecule RNA.

The ribosome reads the code and assembles long polymer chains out of amino acids. These chains then fold to form proteins, and it is the proteins that go on to build and maintain our bodies.

An artificial ribosome, like the one developed in Manchester, functions in a similar way.

The machine's chemical structure is based on a rotaxane, a molecular ring threaded on to a rod. A "reactive arm" is attached to the ring and works its way down the rod, removing and stacking the amino acid units bound to the rod.

A key point is that the sequence for a new polymer chain comes from the scientists. It is controlled by the chemical groups placed along the rod structure.

"The degree of control we have in this is exquisite," said Prof Leigh.

At the moment, the team is only producing small molecules, or peptides, that comprise chains of just a few units in length. This will have to be scaled up to the many tens of units achieved by ribosomes.

But just as insulin for diabetics today is produced in vast vats by engineered microbes, so in the future the Manchester team envisages containers carrying millions upon millions of their artificial machines all churning out the programmed molecules.

"Even though each machine is producing one molecule at a time, if you have a million, million, million of them, all acting to produce the same molecule, then you can produce qualities of the molecule that you can see in a reasonable period of time," said Prof Leigh.
